Where We Think Each Role Lands
Top Lane

Malphite, Nasus and Mordekaiser are all missing from the notes, so the top lane God Tier stays as it is.
Darius gets the biggest top lane buff. The cooldown on Decimate [Q] drops from 5 seconds to 4 at rank five, which means more healing and more bleed stacks in a long fight. He sits in Tier 2 with one of the lower win rates there, though. We have him climbing toward the top of Tier 2 rather than jumping a tier.
Sion gains 95 health per level instead of 87, and Unstoppable Onslaught [R] now triples its damage over the first three seconds of charging, so a full charge hits for up to 1,350 at rank three instead of 1,200. Riot wants him rewarded for driving the ult into a fight instead of using it to run away. He climbs inside Tier 2 as well.
Yone's nerf reaches top too, where he's Tier 1, and it's five points off the attack damage ratio on Mortal Steel [Q]. He holds.
Top lane Varus is the version Riot was aiming at: his W-max builds lose a chunk of on-hit and percent health damage from Blighted Quiver [W]. He was already in Tier 4 and stays there.
Jungle

Cho'Gath and Briar aren't in the notes. Wukong is, and Riot aimed the change at his jungle games. Stone Skin [Passive] gives 4.5 armor per stack at level one instead of 6 (the gap closes by level eighteen), while his base health regeneration rises from 3.5 to 5.5 to help him in lane. It's a small early cut. He keeps his seat.
Lee Sin takes two nerfs at once. Dragon's Rage [R] drops from 175/400/625 base damage to 150/350/550, and Safeguard [W] comes back after 8 seconds instead of 7. Riot said the last Safeguard [W] change gave him too much power. He was already under 50% in Tier 1, and the two cuts together should push him into Tier 2.
That leaves a Tier 1 seat, and Zyra's buff lines up with it. The seeds from Rampant Growth [W] recharge faster through the early and middle ranks, and jungle Zyra already wins plenty of games for a Tier 2 pick. We have her moving up.
Rammus's Defensive Ball Curl [W] now gives 30 to 50% bonus armor and magic resistance instead of 30 to 60%, plus two less flat armor. He has the best win rate of any Tier 1 jungler, so he slides a bit inside Tier 1 rather than out of it.
Nidalee gets less attack speed from Primal Surge [E], and her cougar Pounce [W] hits softer. She was already under a 50% win rate in Tier 2, and she drops to Tier 3. Qiyana's Elemental Wrath [Q] loses part of the damage it gained in 26.17, and that lands on top of the Bastionbreaker cut, an item she finishes in about three jungle games out of four. She's at the bottom of Tier 3 and should fall back to Tier 4.
Xin Zhao and Nunu & Willump both climb inside Tier 3. Xin's Three Talon Strike [Q] scales harder with bonus attack damage, and his attack speed builds get more out of Audacious Charge [E]. Nunu's Absolute Zero [R] hits between 14% and 20% harder depending on rank, which hands back damage he lost when Hextech Rocketbelt was nerfed.
Mid Lane

Vex and Fizz aren't in the notes. Yone is: Mortal Steel [Q] loses five points of attack damage ratio, which Riot calls a moderate nerf after his last changes left him too strong. He's far enough ahead to keep his God Tier spot in mid.
Viktor's cuts are all small and they stack. The shield from Siphon Power [Q] is smaller, scales less with ability power and lasts 2 seconds instead of 2.5, and his base move speed drops from 335 to 330. Riot wants him easier to punish in early skirmishes. He slides a bit down Tier 1 without leaving it.
Taliyah's Unraveled Earth [E] gets ten more base damage and ten more points of ability power ratio at every rank, and she climbs inside Tier 3 (Riot pitched the buff at jungle Taliyah, but mid is where she shows up on our board). Vel'Koz climbs inside Tier 3 as well, with a stronger Organic Deconstruction [Passive] and a one second shorter cooldown on Plasma Fission [Q].
Ryze's Overload [Q] does more bonus damage to Flux-marked targets at its top ranks and gives more move speed, and Rune Prison [W] scales harder with ability power. All of it is aimed at his late game. Orianna gets five more points of ratio on Command: Attack [Q] and Command: Dissonance [W], and Azir gets ten more mana per level. All three are in Tier 4 and stay there.
Twisted Fate's Destiny [R] now reveals untargetable units, which is a bug fix more than a buff. He stays in Tier 2.
Bot Lane

Tristana's Rapid Fire [Q] gives 40% attack speed at rank one instead of 60%, with smaller cuts up to rank four. Riot doesn't like level one Tristana pressing Rapid Fire [Q] and auto attacking her way into control of the lane, and the change is aimed squarely at that window. It's the biggest single hit in bot lane this patch, and she was already the second of our two God Tier bot laners. We have her dropping to Tier 1.
Twitch takes her place. He isn't in the notes at all, and he goes from the top of Tier 2 straight to God Tier. Stealth lets him walk into a fight from an angle nobody is watching, and once Spray and Pray [R] is up he shreds a whole team from behind his frontline. He's also the bot laner most able to leave lane and pick up kills elsewhere on the map.
Miss Fortune climbs to the top of Tier 1. Make It Rain [E] hits harder at every rank, and the waves from Bullet Time [R] now crit for 45% extra damage instead of 30%. She holds Infinity Edge in close to two games out of three, so the crit change reaches most Miss Fortunes. She also holds Hubris in more than half her games, and Hubris now gives ranged champions less attack damage (Riot sized her buffs to cover that). In a teamfight, the bigger Bullet Time [R] is worth more than the attack damage she gives up.
Caitlyn stays in Tier 1, just behind Miss Fortune. Riot moved some of her late game headshot damage into Piltover Peacemaker [Q], which now hits for 230 at rank five instead of 210. She still has the longest base attack range of any marksman and the traps to set up her jungler's ganks, so she keeps winning lanes early.
Jinx loses 20 base health and a quarter point of attack damage per level, about four by level eighteen. Two small cuts. She stays in God Tier (she buys Gluttonous Greaves in about one game in twenty, so the boots change barely reaches her).
Gluttonous Greaves costs 100 more gold. Zeri, Yunara and Samira (all Tier 2) and Kalista (Tier 3) buy it in well over a third of their games, and each of them slides a little inside their tier. Varus's W-max builds lose damage while his Q-max builds gain it. He's Tier 4 and stays there.
Support

Thresh, Leona and Blitzcrank are all absent from the notes. Nothing changes at the top.
Zyra's faster seed recharge helps her in support as much as in the jungle, and she climbs inside Tier 1. Soraka slipped to the top of Tier 2 late in 26.20. She's the one support who already buys Dawncore, in about a quarter of her games, and it now gives 15 ability power for every 100% base mana regeneration instead of 10. That puts her back in Tier 1. Vel'Koz's buffs reach support too, where he climbs inside Tier 2.
Taric's Starlight's Touch [Q], Dazzle [E] and Cosmic Radiance [R] can land on untargetable allies again. It's a bug fix, so he stays in Tier 3.
Everything above was written the day Riot published the 26.21 notes, before anyone had played a ranked game on the patch. The League of Legends tier list hub carries the measured rankings and takes its first read on this patch once the games exist.
